FOXO4-DRI is a senolytic research peptide built with a D-retro-inverso (DRI) modification, which improves its stability and cell permeability.
It targets the interaction between the FOXO4 protein and the tumor suppressor p53. Senescent cells depend on this binding to avoid programmed cell death. By occupying the FOXO4 interface, FOXO4-DRI releases p53, and research has examined how this leads to apoptosis in senescent cells, where FOXO4 is overexpressed.
The peptide gained attention through the work of Baar et al. (Cell, 2017) and has been studied in naturally aged and progeroid mouse models. It is used mainly in cellular senescence research.
